James Clayland was born on 1648 in Gloucester, Gloucestershire, England to Henry Clayland and Anne Coursey Clayland. James married Elizabeth [sic: Penelope] Hemsley on 1673 in Talbot County, Maryland. Penelope Hemsley was born on December 7, 1657 in Sundridge, Kent, England to William Hemsley and Judith (MNU) Hemsley. Their daughter was Judith Clayland, born 26 Dec 1674 in Talbot County.

Rev. James Clayland married (2), Elizabeth, daughter of Roger Nettleship. They had 7 children: William Clayland, Roger Clayland, John Clayland, Catharine Clayland, Rebecca Clayland, and Elizabeth Clayland.

He died on November 7, 1699 in Talbot County, Maryland.
